Lipoprotein DolP supports proper folding of BamA in the bacterial outer membrane promoting fitness upon envelope stress

چکیده

In Proteobacteria, integral outer membrane proteins (OMPs) are crucial for the maintenance of envelope permeability barrier to some antibiotics and detergents. Enterobacteria, stress caused by unfolded OMPs activates sigmaE (? E ) transcriptional response. ? upregulates OMP biogenesis factors, including ?-barrel assembly machinery (BAM) that catalyses folding. Here we report DolP (formerly YraP), a -upregulated poorly understood lipoprotein, is fitness in cells undergo stress. We demonstrate interacts with BAM complex associating membrane-assembled BamA. provide evidence important proper folding BamA overaccumulates membrane, thus supporting integrity. Notably, mid-cell recruitment had been linked regulation septal peptidoglycan remodelling an unknown mechanism. now reveal that, during stress, loses its association mid-cell, thereby suggesting mechanistic link between impaired late step cell division.

Species-Specificity of the BamA Component of the Bacterial Outer Membrane Protein-Assembly Machinery

The BamA protein is the key component of the Bam complex, the assembly machinery for outer membrane proteins (OMP) in gram-negative bacteria. We previously demonstrated that BamA recognizes its OMP substrates in a species-specific manner in vitro. In this work, we further studied species specificity in vivo by testing the functioning of BamA homologs of the proteobacteria Neisseria meningitidis...

BamA Alone Accelerates Outer Membrane Protein Folding In Vitro through a Catalytic Mechanism

β-Barrel assembly machinery protein A (BamA) plays a critical role in the biogenesis of outer membrane proteins (OMPs); however, a mechanistic understanding of its function is lacking. Here, we report an in vitro assay that investigates whether the mechanism of BamA-catalyzed OMP folding is stoichiometric or catalytic. We found that BamA accelerates the folding of OMPs in vitro via a catalytic ...

Interaction of an autotransporter passenger domain with BamA during its translocation across the bacterial outer membrane.

Autotransporters are a superfamily of virulence factors produced by Gram-negative bacteria consisting of a large N-terminal extracellular domain ("passenger domain") and a C-terminal beta barrel domain ("beta domain"). The mechanism by which the passenger domain is translocated across the outer membrane (OM) is unknown. Here we show that the insertion of a small linker into the passenger domain...

Outer membrane β-barrel protein folding is physically controlled by periplasmic lipid head groups and BamA.

Outer membrane β-barrel proteins (OMPs) are crucial for numerous cellular processes in prokaryotes and eukaryotes. Despite extensive studies on OMP biogenesis, it is unclear why OMPs require assembly machineries to fold into their native outer membranes, as they are capable of folding quickly and efficiently through an intrinsic folding pathway in vitro. By investigating the folding of several ...

Conformational Plasticity of the POTRA 5 Domain in the Outer Membrane Protein Assembly Factor BamA.

BamA is the main component of the β-barrel assembly machinery (BAM) that folds and inserts outer membrane proteins in Gram-negative bacteria. Crystal structures have suggested that this process involves conformational changes in the transmembrane β-barrel of BamA that allow for lateral opening, as well as large overall rearrangements of its periplasmic POTRA domains. Here, we identify local dyn...
